How do you make sodium citrate anticoagulant solution?

250 mL SODIUM CITRATE 4% W/V ANTICOAGULANT SOLUTION USP is a sterile solution in a PVC bag. Each 100 mL contains: Sodium Citrate (dihydrate) 4.0 g; and 100 mL Water for Injection, (pH adjusted with citric acid). DO NOT INFUSE SODIUM CITRATE 4% W/V ANTICOAGULANT SOLUTION USP DIRECTLY TO THE PATIENTS.

How sodium citrate works as an anticoagulant?

SODIUM CITRATE 4% W/V ANTICOAGULANT SOLUTION USP acts as an extracorporeal anticoagulant by binding the free calcium in the blood. Calcium is a necessary co-factor to several steps in the clotting cascade. The following ingredients are key components of the solution: Citric acid for pH regulation.

Why do we use sodium citrate for coagulation tests?

Background :Sodium citrate has been used as a coagulation test because factor V and VIII are more stable in a citrated specimen. Ethylenediaminetetraacetic acid (EDTA) has been used for the hematologic test because blood cells are preserved better in the EDTA specimen.

How do you make anticoagulant blood?

Here is a quick protocol to make ACD anticoagulant.

  1. Dissolve 1.32g of sodium citrate in 85ml of distilled water.
  2. Dissolve 0.48g of citric acid in the solution from step 1.
  3. Dissolve 1.47g of dextrose in the solution from step 2.
  4. Add distilled water to 100ml.
  5. Filter sterilize through 0.2um filter.

How do you make 1 sodium citrate?

Make a solution of 1% sodium citrate (0.5 g in 50 mL of H2O). Heat the 125 mL solution of AgNO3 until it begins to boil. Add 5 mL of 1% sodium citrate solution, as soon as boiling commences.

How does EDTA work as an anticoagulant?

The mechanism of EDTA anticoagulant action is based on inhibition of thrombocyte aggregation and various reactions of hemostatic cascade due to chelation of free Ca2+ ions. Blood cells of various animals show different reactions to various anticoagulants.

What is citrate anticoagulant?

Citrate is essentially a regional extracorporeal anticoagulant, with a short systemic half-life of around 5 min, metabolized predominantly by mitochondria in the liver, skeletal muscle and the kidney.

How do you make EDTA anticoagulant?

Anticoagulant EDTA 0.4 M at pH about 7.4 to 8.0: dissolve 150 g disodium EDTA dihydrate in 700 ml dH2O. Adjust pH to 8.0 adding slowly about 40.3 ml of 10 M NaOH, fill up to 1 liter with dH2O and filter to sterilize.

How is EDTA used as an anticoagulant?

Ethylenediaminetetraacetic acid (EDTA) is a well-known anticoagulant since early 1950s and it has certain advantages over other anticoagulants [2]. It inhibits clotting by removing or chelating calcium from the blood.

How do you make 2% sodium citrate?

For every 2.1 g of citric acid, use 2.5 g of sodium bicarbonate and if everything goes well, you’ll get 2.9 g of sodium citrate after evaporating all the water. Just mix the two ingredients with which ever liquid you’re using (milk or water) and you should be good to go, clearly no need to evaporate the water.

Is sodium citrate an anticoagulant?

Background: Sodium citrate has been used as an anticoagulant to stabilize blood and blood products for over 100 years, presumably by sequestering Ca(++) ions in vitro.

Is sodium heparin an anticoagulant?

In other words, sodium heparin, an anticoagulant used widely for blood collection, has been known to inhibit DNA polymerase activity in PCR assays (2).

What is in a sodium citrate anticoagulant solution?

Sodium Citrate Anticoagulant Solution Description. Each 100 mL of SODIUM CITRATE 4% W/V ANTICOAGULANT SOLUTION USP contains: Sodium Citrate (dihydrate) 4.0 g; and 100mL Water for Injection (pH adjusted with citric acid). The PVC bag is not made with natural rubber latex. The bag is made from a multilayered film.

Why is sodium citrate used as an additive in a defibrillator?

When the blood is not simply defibrinated, sodium citrate is the additive most commonly used.[n2] [n3] The citrate ion chelates calcium ions in the blood by forming calcium citrate complexes, disrupting the blood clotting mechanism.

What are the warnings and precautions for sodium citrate?

Warnings and Precautions 1 Verify that the SODIUM CITRATE 4% W/V ANTICOAGULANT SOLUTION USP has been securely attached to the anticoagulant (AC) line on the system tubing set. 2 Single-use container. Do not reuse. Discard any unused or partially used product. 3 Rx only.

How much sodium citrate do you put in a blood test?

There’s a lot of variation in how much sodium citrate solution you should use to achieve the best results. Too little and the blood might clot, too much and the tart taste will be overwhelming. Overall, the average tends to be around 25-70 ml of a 3.8% solution for A liter of blood.
